Lethality acts in the 2nd/early 3rd instar. Number of boutons on embryonic somatic muscles is 50% wild type, though muscle surface area is normal. The size of the boutons is abnormally large, such that synaptic surface area for all boutons on a muscle is similar to that of wild type. Microtubules in boutons are disorganised, and punctae are evident. Localization of syt, Csp, Fas2 and dlg1 proteins is normal, indicating normal axonal transport.

Expression of Hsap\VAPBScer\UAS.cCa in a Vap-33-1Δ448 mutant background, under the control of Scer\GAL4C164 or Scer\GAL4D42 rescues the larval lethality. Rescued flies are fertile and do not show any obvious morphological or behavioral defects.
In synapses expressing Hsap\VAPBScer\UAS.cCa under the control of Scer\GAL4C164 or Scer\GAL4D42 in a Vap-33-1Δ448 background, the number of synaptic boutons is similar to controls. Electrophysiological analysis of the same synapses shows that the EJPs and mEJPs are both similar to controls. Hence, the lethality, aberrant NMJ morphology and increased mEJP amplitude associated with loss of Vap-33-1 are rescued by Hsap\VAPBScer\UAS.cCa neuronal expression.
Vap-33-1Δ448 mutant lethality is rescued through expression of Hsap\VAPBP56S.Scer\UAS under the control of either Scer\GAL4C164 or Scer\GAL4D42. However, the number of boutons is not significantly different in flies expressing Hsap\VAPBP56S.Scer\UAS compared to controls. Moreover, no significant difference in EJPs or quantal sizes is observed in flies expressing Hsap\VAPBP56S.Scer\UAS.
The rough eye phenotype caused by Hsap\MAPTGMR.Ex.PJ is suppressed by Vap-33-1Δ448/+.
