In larvae in which scribScer\UAS.cBa expression is driven by either Scer\GAL4BG487 or Scer\GAL4C380, the number of active zones is significantly higher than in wild-type boutons. In larvae in which scribScer\UAS.cBa expression is driven by either Scer\GAL4BG487 or Scer\GAL4C380, the density of synaptic vesicles is significantly lower than that observed in wild-type boutons, indicating a gain-of function. Presynaptic expression of transgenic scribScer\UAS.cBa under the regulation of Scer\GAL4C380 results in a significant increase in the number of synaptic active zones compared to wild-type.

Expression of scribScer\UAS.cBa under the control of Scer\GAL469B fully rescues the cell polarization and disc overgrowth phenotypes of scrib673/scribird15 transheterozygotes.

In larvae in which scribScer\UAS.cBa expression is driven by either Scer\GAL4BG487 or Scer\GAL4C380, the density of synaptic vesicles is significantly decreased from the levels found in scrib673 mutants. In larvae in which scribScer\UAS.cBa expression is driven by either Scer\GAL4BG487 or Scer\GAL4C380, the number of active zones is significantly higher than in both scrib1 and scrib673 mutants and wild-type boutons. In contrast, the defect in the basal lamina is only partially rescued by transgenic scribScer\UAS.cBa expression. Presynaptic expression of transgenic scribScer\UAS.cBa under the regulation of Scer\GAL4C380 in a scrib673 background rescues the decrease in miniature excitatory junctional currents compared with wild-type. Expression of scribScer\UAS.cBa under the regulation of Scer\GAL4C380 not only rescues the decrease in active zone number found in scrib673 mutants but results in a significant increase in the number of active zones compared to wild-type. Presynaptic expression of transgenic scribScer\UAS.cBa under the regulation of Scer\GAL4C380 in a scrib673/Df(3R)Tl-X background has no effect on the miniature excitatory junctional currents (mEJC), indicating that the reduced mEJC amplitude in scrib673/Df(3R)Tl-X mutants arises from a postsynaptic mechanism.
