The number of nuclear bundles and individualization complexes is significantly decreased in the testis of ctpDIIA82 mutants compared to controls. The F-actin is decreased in the investment cones during ctpDIIA82 sperm individualization compared to wild-type controls. This phenotype is not temperature sensitive.

ctpDIIA82 homozygotes and the following transheterozygotes exhibit thin bristles and erect wings: ctpDIIA82/ctpDIIB14, ctpDIIA82/ctpexc41, ctpDIIA82/ctpDIA5, ctpDIIA82/ctpins1, ctpexc6/ctpDIIA82 and ctpDIIA82/Df(1)bi-DL1. The seminal vesicles of ctpDIIA82 hemizygous males contain drastically lower numbers of motile sperm than wild type. Any motile sperm are also more sluggish than in wild type. The testes of 2 day-old ctpDIIA82 mutants contain fewer cysts than wild type; the mutant cysts appear to be shorter and contain contain spermatids that are abnormally bundled in multiple fascicles within a single cyst. There are very few elongation cones (ECs) in the testis of ctpDIIA82 males and those present are deformed. The average length of cysts is shortened in ctpins1 testes, as the peak of the elongation cone (EC) distribution profile is shifted to the basal end. The ctpDIIA82 mutation affects the cellularization of spermatids. Mature cysts show incompletely individualized spermatids with multiple axoneme-mitochondria sets within one cellular envelope that are interconnected by thin membranous bridges. The organization of axonemal microtubules appears to be normal.

Almost all ctpins1; sw1/Y males fail to emerge, at either 25oC or 18oC.

The F-actin density in the investment cones of spermatid individualization complexes is further decreased following a heat pulse of 11 hours in ctpDIIA82, shi2 double mutants compared to shi2 single mutants. The same phenotype is enhanced in ctpDIIA82, shi1 double mutants compared to shi1 single mutants.

The ctpDIIA82 mutation is lethal in combination with one copy of Gl1. The sterility of ctpDIIA82 males is partially suppressed when they carry one copy of Dhc64C4-19.

Expression of Hsap\DNCL1Scer\UAS.T:Hsap\MYC, under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, rescues the reduced fertility phenotype of ctpDIIA82/Y males. This rescue includes increasing the amount of motile sperm within the seminal vesicles and increasing the amount and length of ECs.
