Testes of homozygous Rexo5c04255 second instar male larvae are much smaller in size compared to age-matched controls and their gonioblast divisions are arrested at the first or second mitotic division, assed by the level of fusome branching.

Rexo5c04255 mutants are rescued to adulthood by Scer\GAL4Act.PU-driven expression of Rexo5Scer\UAS, yet (due to poor expression in germline) the males display defects in differentiation of the spermatogonial cells as well as distribution of multicellular germ cell cysts that are now present throughout the testicular tube (rather than being restricted to the apical tip as in wild-type); no meiotic nuclei are detected in the mutant germ cells at day 3 post-eclosion. The rescued females are sterile with only rudimentarily developed small ovaries (reminiscent of the string-of-pearls phenotype), the oocytes do not fully develop and display egg-chamber degeneration at stages 4-6. Near complete loss of germline stem cells and germarium proliferation defects are observed at 14 days post-eclosion.

The second larval instar lethality of Rexo5c04255 homozygotes is rescued by combination with either Rexo5+t4.5 or Rexo5+t6.5 or by expression of Rexo5Scer\UAS driven by Scer\GAL4Act.PU. Partial rescue (the larvae survive until the third instar stage) is achieved by expression of Rexo5H493R.Scer\UAS, but no rescue is effected by expression of any of the following: Rexo5D361A.Scer\UAS, Rexo5E363A.Scer\UAS, Rexo5H493A.Scer\UAS, Rexo5D446A.Scer\UAS, Rexo5D361A.D446A.Scer\UAS or Rexo5E363A.D446A.Scer\UAS (all tested with the Scer\GAL4Act.PU driver).

Crossing Rexo5c04255 to Df(3L)BSC411 is lethal while Rexo5c04255/Df(3L)BSC410 flies are viable.
