mague00439 homozygotes and mague00439/maguf02256 transheterozygotes are viable and exhibit a wing vein defect.

mague00439/maguf02256 transheterozygous testes contain fewer germ cells. The median germline stem cell number per testis is only 3, whereas sibling controls carry a median of 8 germline stem cells at 3rd instar larval stage. Moreover, mutant testes display germ cells with branched fusomes next to the hub, indicating that they have differentiated and are no longer bona fide stem cells.

Expression of tkvQ199D.Scer\UAS.T:Ivir\HA1 using the germ cell driver Scer\GAL4nos.UTR.T:Hsim\VP16 raises the fraction of testes with germline stem cells from 63% to 100% in a mague00439/maguf02256 background. The median germline stem cell number also doubles compared to that observed in mutants.

Expression of maguScer\UAS.T:SV5\V5 under the control of Scer\GAL4upd in mague00439/maguf02256 flies aged at 29[o]C partially rescues the number of germline stem cells. The fraction of testes with germline stem cells increases to more than 50% and sometimes approaches or equals 100%. Partial rescue occurs in both flies aged 3 days and 12 days. A similar rescue is displayed for the wing vein phenotype.
