    Detailed Description

In sdkMB05054 stage 7-8 embryos, cells of the dorsal epidermis exhibit less straight boundaries (visualized by Ecad-GFP) than controls.

Laser ablation experiments in sdkMB05054 stage 10-11 embryos show that the bicellular bonds in the epidermis (visualized by Ecad-GFP) are under lower tension than controls.

Stage 15-16 sdkMB05054 homozygous embryos exhibit incomplete tracheal cell intercalation (many tracheal branches still maintain intercellular junctions, visualized by Ecad staining) different than controls.

In sdkMB05054 stage 17 embryos the epithelial permeability barrier is intact as in controls.

In sdkMB05054 pupae the ommatidial arrangement of retinal pigment cells is frequently irregular (visualized by Ecad staining); there are some extra cells within the lattice, while at other positions cells are missing or bristles are misplaced when compared to controls. Small sdkMB05054 mutant clones also exhibit extra or missing pigments cells, indicating a cell-autonomous effect, as well as missing bristles, as compared to controls.

sdkMB05054 male pupal A8a epithelia cells take longer to resolve four-way vertices and show decreased junction growth rate, but show no changes in junction shrinkage rate, as compared to controls. A small proportion of adult males exhibit mis-oriented genitalia.

The germ band of sdkMB05054 seems to have tighter cell-cell contacts, as indicated by the decreased levels of dextran signal at cell junctions upon injection into the perivitelline space, as compared to controls.

sdkMB05054 homozygous adults tethered to a trackball fail to follow the motion of square wave contrast patterns with short wavelengths, but not with medium or long wavelengths, as compared o controls.

sdkMB05054 homozygous adults display significantly more photoreceptor R1-R6 axons overshooting the lamina and terminating in the medulla, despite most axons targeting the correct optic neuropil, and display defective alignment of lamina neurons along R1-R6 axons with some neurons locating beneath the lamina plexus, despite most lamina neurons still being present, as compared to heterozygous controls. Mutant adults also show defective neural superposition, with cartridges frequently frequently fused together or receiving too many or too few photoreceptor axons, despite of no significant changes in the average number of photoreceptors per ommatidium, as compared to controls; cartridge organization is also defective within sdkMB05054 homozygous clones. Mutant pupae also show irregular projection of the photoreceptor R4 axons within the pupal lamina, namely irregular distance and direction of extension from the ommatidium instead of their consistent direction and direction in controls.

Stage 15-16 sdkMB05054 cnoR2 and sdkMB05054 Df(3R)pydB12 double heterozygous embryos exhibit incomplete tracheal cell intercalation (many tracheal branches still maintain intercellular junctions) similar to sdkMB05054 homozygotes and different than controls.
