UASt regulatory sequences drive expression of an inverted repeat.
Expression of Unc-13-4BGD7394 (along with UAS-Dcr-2 expression) driven by Scer\GAL4phm.PO leads to developmental delay/arrest at the larval stage (around 3% at L1/2 and 8% at L3). Arrested third instar larvae also show an overgrowth phenotype due to the extended larval feeding period. Those larvae that do initiate pupariation are significantly delayed in doing so and exhibit overgrowth phenotypes. Feeding ecdysone (E) to larvae almost fully rescues the phenotypes.
Adults expressing Unc-13-4BGD7394 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
