Ca-α1TGal4 heterozygous adults expressing Ca-α1TGD3252 under the control of Scer\GAL4Ca-α1T (the intrinsic Gal4 driver of Ca-α1TGal4), previously trained under 12h:12h light:dark cycles, exhibit a significant decrease in sleep time when kept under the same light:dark cycles, but exhibit a significant increase in sleep time after being kept for 48h-96h under constant darkness, as compared to controls.

The expression of Ca-α1TGD3252 under the combined control of Scer\GAL4elav.Switch.PO and treatment with RU486, results in adults previously trained under 12h:12h light:dark cycles exhibiting a significant increase in sleep time during the subjective light period after being transferred into constant darkness conditions, as compared to controls; expression under the control of Scer\GAL4elav-C155 results in adults exhibiting a significant increase in sleep time, regardless of individuals being kept under 12h:12h light:dark cycles or under constant darkness, as compared to controls; expression under the control of Scer\GAL4104Y results in adults exhibiting a significant decrease in sleep time under 12h:12h light:dark cycles, but not under constant darkness, as compared to controls; no significant changes in overall sleep time are observed upon expression under the control of Scer\GAL4VGlut-OK371, Scer\GAL4Gad1.PU, Scer\GAL4ChAT.7.4, Scer\GAL4ple.PF, Scer\GAL4Tdc2.PU, Scer\GAL4l.Trh.PK, Scer\GAL4386Y, Scer\GAL4Ilp2.PR, Scer\GAL4GMR.PF, Scer\GAL4210Y, Scer\GAL4smid-C161, Scer\GAL4Alp4-c232, Scer\GAL4c465, Scer\GAL4c309, Scer\GAL4Mef2.247, Scer\GAL4Clk.8.0, Scer\GAL4dimm-929, or Scer\GAL4P2.4.Pdf.

Adults expressing Ca-α1TGD3252 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in early pupal lethality.
