Expression of Rae1GD14705 under the control of Scer\GAL4bam.T:Hsim\VP16 (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi) results in male sterility. Primary spermatocytes contain nuclear herniations, onion stage spermatids have nuclei and Nebenkern of varying sizes and spermatid bundles contain round, scattered and irregularly sized nuclei. Meiotic defects are seen, including irregular metaphase plates in metaphase I and lagging chromosomes and unbalanced chromosome segregation in anaphase I.
Brain neuroblasts of third instar larvae expressing Rae1GD14705 under the control of Scer\GAL4wor.PA have a significantly higher mitotic index compared to wild type. 70% of anaphase figures have defects, including lagging chromosomes, chromatin bridges and uneven chromosome segregation.
Adults expressing Rae1GD14705 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
