As in wild type, Or67dGAL4-1 mutant males exhibit inhibition of female contact-induced male-male aggression seen in wild type males that have previously been exposed to females.
Naive mutant males court virgin and mated males equally and do not benefit from prior training.
Or67dGAL4-1 males show relatively wild-type levels of aggression to male flies in which the hydrocarbon-producing oenocytes have been eliminated (through expression of WScer\UAS.cZa, and Avic\GFPStinger.Scer\UAS.T:nls-tra under the control of Scer\GAL4desat1.PB and Scer\GAL80ts.αTub84B at 18[o]C during development and 25[o]C at the adult stage).
The courtship index of Or67dGAL4-1 males paired with wildtype females is not significantly different from wild-type controls. Addition of 11-cis-vaccenyl acetate (cVa) to the mating chamber fails to suppress courtship in mutant males towards wild-type females. Indeed, the courtship index of these males in the presence of cVa is significantly higher than in wild-type male controls.
Synthetic 11-cis-vaccenyl acetate (cVA) does not promote male-male aggression in Or67dGAL4-1 males, in contrast to wild-type males.
Or67dGAL4-1 males paired with wild-type males show a significantly higher male-male courtship index than that of control males paired with wild-type males.
Or67dGAL4-1 females mated to wild-type males show a higher copulation latency than control females mated to wild-type males.
The male pheromone cVA ([Z]-11-octadecenyl acetate) elicits a rapid and robust firing response in the T1 trichoid sensilla of control Or67d+1 flies but not in those of Or67dGAL4-1 flies. Male and female Or67dGAL4-1 flies do not produce a detectable electroretinogram response to cVA (in contrast to controls), but they show a normal electroretinogram response to ethanol.
When paired with virgin females, Or67dGAL4-1 males court at levels comparable to those of control males. However, when Or67dGAL4-1 males are paired with wild-type males, they show a roughly threefold higher courtship activity than control males paired with wild-type males.
Or67dGAL4-1 females mated to wild-type males show a higher latency to copulation than control females mated to wild-type males.
Virgin Or67dGAL4-1 females are courted as vigorously as control females when paired with wild-type males.
Application of the male pheromone cVA ([Z]-11-octadecenyl acetate) to the abdomens of virgin females suppresses courtship by control males but not by Or67dGAL4-1 males in a single-pair courtship assay.
Or67dGAL4-1 is a suppressor of abnormal courtship behavior | heat sensitive phenotype of Scer\GAL4ple.PF, TrpA1UAS.(B).cKa
Or67dGAL4-1 is a suppressor of abnormal neurophysiology phenotype of lusht.D118A
Or67dGAL4-1 is a suppressor of abnormal courtship behavior phenotype of lusht.D118A
The suppression of courtship towards mated females which is seen in males expressing TrpA1Scer\UAS.(B).cKa under the control of Scer\GAL4ple.PF which have been shifted to 30[o]C for 45 minutes and subsequently returned to 25[o]C is not seen in a Or67dGAL4-1 background.
The lusht.D118A-induced activation of T1 neurons is mediated by Or67d as an Or67dGAL4-1 background abolishes this activating effect.
A Or67dGAL4-1 background completely reverses the lusht.D118A-induced suppression of male courtship, indicating these behavioral changes are mediated through T1 neurons.
A Or67dGAL4-1 background completely suppresses the increase in courtship behaviour seen in lusht.D118A females.
