ScerGAL45-HT2-PL00052 is expressed in commissures connecting the left and right hemispheres of the early third instar larval brain, in clusters of cells within the medial regions of the hemispheres, a small population of cells within the developing optic ganglia, and in a small number of cells in the ventral ganglia. In the pupal brain, the larval expression pattern of ScerGAL45-HT2-PL00052 is replaced by expression in a disc of cells at the base of the developing optic ganglia and in cells that may be glia. In the adults, expression of ScerGAL45-HT2-PL00052 is seen in cells in the protocerebrum that include multipolar neurons around the calyces. It is also expressed in gustatory neuron that project into the subesophageal ganglion.
Ablation (using hidUAS.cUa and rprUAS.cUa) of Scer\GAL45-HT2A-PL00052 cells significantly reduces aversive olfactory memory in larvae trained using salt, but not electric shocks, compared to controls, with no effect on naive preference for olfactory or gustatory stimuli and no effect on appetitive (fructose) olfactory memory.
Acute inhibition (using shi1.UAS at 35[o]C) of Scer\GAL45-HT2A-PL00052 cells in larvae during training and testing does not affect aversive (salt) olfactory memory or naive preference for olfactory or gustatory stimuli, compared to controls.
