    Detailed Description

    Reference

Electrical sound responses of Johnston's organ neurons are virtually abolished in Dnai2MB06262 mutant flies, although some residual responses to intense sound stimuli are retained. Mechanical amplification is also completely abolished.

No gross morphological defects are seen in the Johnston's organ neurons in Dnai2MB06262 mutant flies but the outer dynein arms in the proximal region of the cilium are selectively lost.

Homozygous Dnah3MB05004 mutant males are fertile and sperm appear motile. There is a significant reduction in sperm competition compared with controls.

Compound action potentials can be evoked by sound in the antennal nerve of mutant flies, but the sound particle velocities required to elicit the response is increased compared to wild type. The displacement response of the antenna over a range of sound particle velocities is linearised, indicating loss of mechanical amplification.

As in either mutant alone, iav1 Dnai2MB06262 double mutants entirely lack sound-evoked electrical potentials. The excessive mechanical amplification seen in single iav1 mutants is entirely abolished in the double mutants, as in single Dnai2MB06262 mutants.

Expression of Dnai2MB06262 rescues the sound-evoked electrical and mechanical response defects seen in Dnai2+t20 mutant flies.

Expression of Dnai2Scer\UAS.T:Avic\GFP-YFP in chordotonal neurons under the control of Scer\GAL4nan.PK rescues the sound-evoked electrical and mechanical response defects seen in Dnai2MB06262 mutant flies.

Precise excision of the Mi{ET1}Dnai2MB06262 element restores the sound-evoked electrical and mechanical response defects seen in Dnai2MB06262 mutant flies. The loss of the outer dynein arms in the ciliary axonemes of Johnston's organ neurons is also restored.
