Description

A synthetic oligo (CTAGTCTAGACTAG) has introduced stop codons in all 3 reading frames at +502 bp relative to the common transcription initiation site of Tgs1 and moi. This introduces a stop codon into moi, but does not affect the sequence of Tgs1.

P{lacW}moi^S096713/Df(3R)d189 animals carrying P{CaSpeR-DTLu^m} (i.e. moi mutants) eclose and survive for a short time, exhibiting signs of locomotor defects, rough eyes, missing wing veins, scalloped wings and missing thoracic bristles.

Df(3R)d58/Df(3R)d189 animals also carrying P{CaSpeR-DTLu^m} (i.e. moi mutants) have larval neuroblasts whose most prominent cytological defects are chromosome end-to-end associations involving one, two or more chromosomes; cells containing several circular chromosomes; fused chromosomes that form chromosome chains; and telomere associations.

Df(3R)d192/Df(3R)d189 or P{lacW}moi^S096713/Df(3R)d189 neuroblasts that carry P{CaSpeR-DTLu^m} (i.e. moi mutants) exhibit anaphase chromosome bridges.
