UASt regulatory sequences drive expression of an inverted repeat.
Larvae expressing ChroKK105370 under the control of Scer\GAL4insc-Mz1407 display central brain neural stem cell (NCS) reactivation defects: in contrast to wild-type, in the knockdown larvae at 96h after larval hatching (ALH) almost a third of the mira-positive central brain neural NSCs still extend their processes and only 1% of the dpn-positive cells are actively dividing. Expression of ChroKK105370 for two days after all the neuroblast have already reactivated (using tub-gal80[ts] to limit the time of expression) is sufficient to induce re-entry to the quiescent state in vast majority of NSCs.
Scer\GAL4insc-Mz1407-driven expression of ChroKK105370 does not affect NCS size or specification and neither does it induce excessive apoptosis or premature differentiation in the central brain neuroblasts. However, in contrast to controls, almost half of the neuroblasts show only one centriole, suggesting that they are in either G0 or early G1 stage of the cell cycle.
No defect in exiting the quiescent state is observed in the mushroom body neuroblasts at 12h ALH (as in wild-type controls, the cells are mitotically active and do not have extensions) and at 36h ALH, just as in controls, they continue to proliferate in the absence of dietary amino acids. There are also no indications of any cell cycle defects in these neuroblasts.
Scer\GAL4ey-OK107-driven expression of ChroKK105370 leads to formation of shorter mitotic spindle in the mushroom body neuroblasts, but none of these display cellular mira-positive extensions and all are actively dividing cells. Expression controlled by any of the following drivers: Scer\GAL4repo, Scer\GAL4Cg.PA or Scer\GAL4Lsp2.PH does not lead to any neuroblast reactivation defects.
Expression of ChroKK105370 under the control of Scer\GAL4Bx-MS1096 results in wings which are reduced in size, severely blistered and misshapen.

The reactivation defects in central brain neuroblasts (high proportion of the cells retain the cellular extensions, one of the hallmarks of the quiescent state) observed in third instar larvae expressing ChroKK105370 under the control of Scer\GAL4insc-Mz1407 cannot be rescued by co-expression of either CycBScer\UAS.cWa or CycEScer\UAS.cUa.
The ability of Scer\GAL4insc-Mz1407-driven expression of any of the following: InRA1325D.Scer\UAS, RhebScer\UAS.cPa or Pi3K92EScer\UAS.T:Hsap\MYC,T:Hsap\CAAX to induce central brain neuroblast reactivation in the absence of dietary amino acids is prevented by co-expression of ChroKK105370.
