Comments

50-90% of mtg1 homozygous embryos are rescued to hatch by expression of mtgScer\UAS.T:Avic\GFP under the control of Scer\GAL4da.G32. The rescued larvae show coordinated locomotory movement and have a normal anatomy, including rescue of the cuticle defect seen in mtg1 embryos. The majority of the rescued animals die in larval stages. On average, 50% of rescued animals that survive to the third larval instar ae fully rescued to adulthood, and overall up to 20% of mtg1 homozygotes are completely rescued to adulthood by expression of mtgScer\UAS.T:Avic\GFP under the control of Scer\GAL4da.G32.

Expression of mtgScer\UAS.T:Avic\GFP under the control of Scer\GAL4elav-C155 does not rescue viability in mtg1 homozygotes.

Expression of mtgScer\UAS.T:Avic\GFP under the simultaneous control of both Scer\GAL4da.G32 and Scer\GAL4elav-C155 increases full adult rescue of mtg1 homozygotes by more than 3-fold at 18[o]C, compared to expression of mtgScer\UAS.T:Avic\GFP under the control of Scer\GAL4da.G32 alone.

Expression of mtgScer\UAS.T:Avic\GFP under the control of Scer\GAL4how-24B results in 18-32% hatching of mtg1 homozygotes, however, all these larvae die within a few minutes of hatching. The cuticle herniation defect of mtg1 embryos is rescued in these animals.
