UAS regulatory sequences drive expression of a C-terminal part of pros containing an expanded 127Q polyQ tract (a 20Q tract is found in the wild-type protein). The polyQ tract is flanked by 33 amino acids on the N-terminal side and by 369 amino acids on the C-terminal side. The protein is tagged at the C-terminal end with Tag:HA.
Expression of prosZzzz\CAG.127Q.Scer\UAS.T:Ivir\HA1 in the developing eye under the control of Scer\GAL4GMR.PF results in disruption of eye structure and a lack of pigmentation (although the toxicity is less than in Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1 mutants).
Scer\GAL4GMR.PF, pros127Q.UAS.Tag:HA has eye phenotype, non-suppressible by MlfUAS.cKa, Scer\GAL4GMR.PF
Scer\GAL4GMR.PF, pros127Q.UAS.Tag:HA has eye phenotype, non-suppressible by Ecol\lacZUAS.Tag:NLS(SV40-largeT), Scer\GAL4GMR.PF
Scer\GAL4GMR.PF, pros127Q.UAS.Tag:HA has eye phenotype, non-suppressible by Ecol\lacZUAS.cBa, Scer\GAL4GMR.PF
MlfUAS.cKa, Scer\GAL4GMR.PF, pros127Q.UAS.Tag:HA has eye phenotype
Retina expressing MlfScer\UAS.cKa and prosZzzz\CAG.127Q.Scer\UAS.T:Ivir\HA1 both under the control of Scer\GAL4GMR.PF results in retinas that are thinner than wild-type and exhibit aberrant tissue morphology.
The presence of one or two copies of MlfScer\UAS.cKa (with the Scer\GAL4GMR.PF driver) fails to suppress the deterioration of eye structure after 1 day post eclosion found in flies expressing prosZzzz\CAG.127Q.Scer\UAS.T:Ivir\HA1.
Retina expressing Ecol\lacZScer\UAS.T:SV40\nls2 or Ecol\lacZScer\UAS.cBa with prosZzzz\CAG.127Q.Scer\UAS.T:Ivir\HA1, under the control of Scer\GAL4GMR.PF does not suppress the toxic effects of prosZzzz\CAG.127Q.Scer\UAS.T:Ivir\HA1 expression.
