Round developing spermatids are present in testes of His3.3A2X1, His3.3B0 double mutant males but vary in size. Late elongating nuclei are never observed in the double mutants. Clear defects in meiosis are apparent by metaphase I, with diffuse chromosomes that are only partially condensed. Metaphase I with incompletely condensed chromosomes is followed by an aberrant anaphase in the double mutants. Further meiotic aberrations occur in anaphase II in His3.3A2X1, His3.3B0 double mutant testes, such as chromosome bridges between sister chromatids. Compared to wild-types, His3.3A2X1, His3.3B0 double mutant testes contain postmeiotic nuclei that are abnormal in size. The postmeiotic nuclear body is often fragmented into two-fifteen smaller bodies in the double mutants.
His3.3A2X1, His3.3B0 double mutant males also carrying a His3.3AT:Zzzz\FLAG.cSa transgene are viable and fertile. The His3.3AT:Zzzz\FLAG.cSa rescue construct restores all aspects of normal germ lines (meiotic segregation, the single nuclear body in postmeiotic nuclei, production of motile sperm and male fertility) to double mutant males.
His3.3A2X1, His3.3B0 double mutant males also carrying a His3.3AH3.T:Zzzz\FLAG transgene are viable, but sterile. Meiotic defects and fragmentation of the postmeiotic nuclear body persists in His3.3A2X1, His3.3B0 double mutant males carrying His3.3AH3.T:Zzzz\FLAG.
His3.3A2X1, His3.3B0 double mutant males also carrying a His3.3AK4R.T:Zzzz\FLAG transgene are fertile, but they show reduced viability. The His3.3A2X1, His3.3B0 double mutant germ line cells also carrying His3.3AK4R.T:Zzzz\FLAG show no defects.
His3.3A2X1, His3.3B0 double mutant males also carrying a His3.3AK9R.T:Zzzz\FLAG transgene are fertile, but they show reduced viability.
