UAS regulatory sequences drive expression of Hsap\MAPT, carrying mutations in fourteen disease-associated Ser-Thr-Pro sites (Thr 111, Thr 153, Ser 175, Thr 181, Ser 199, Ser 202, Thr 205, Thr 212, Thr 217, Thr 231, Ser 235, Ser 396, Ser 404, and Ser 422) to alanine.

Flies expressing Hsap\MAPTAP.Scer\UAS under the control of Scer\GAL4ple.PF show a significant loss of PPL1 dopaminergic neurons in the brain by 2 weeks after eclosion, with the phenotype becoming more severe as the flies age.

Overexpression of Hsap\MAPTAP.UAS under the control of Scer\GAL4RapGAP1-OK6 in larval segmental nerves shows no significant reduction in MT density; a significant decrease in the number of mobile vesicles with in axons; effects synaptic vesicle kinetics (there is a significant increase in synaptic vesicle pausing time, a significant decrease in net distance run per sec, and a significant decrease in anterograde vesicular transport) as compared to WT controls. Emerging flies have unexpanded wings.

Expression of Hsap\MAPTAP.Scer\UAS under the control of Scer\GAL4GMR.PF is not associated with toxicity in Drosophila.

Compared to controls, expression of Hsap\MAPTAP.Scer\UAS under the control of Scer\GAL4elav.PU does not result in mushroom body abnormalities.

Expression of Hsap\MAPTAP.Scer\UAS in the developing eye under the control of Scer\GAL4GMR.PF generates a mild rough eye phenotype.

Expression of Hsap\MAPTAP.Scer\UAS in the developing external sensory organs under the control of Scer\GAL4Eq1 generates a mild degree of bristle loss.
