Adulthood-only expression of Atg7HMS01358 under the control of Scer\GAL4esg.PU leads to a significant increase in the number of mitotic cells (phospho-H3 staining) in the adult midgut, as compared to controls. (The temporal regulation of expression is dependent on Gal80[ts] and induced by temperature shift to 29[o]C for 5-6 days during adulthood).
Adulthood-only expression of Atg7HMS01358 under the combined control of Scer\GAL4Myo31DF-NP0001 and Gal80[ts] has no obvious effect on the posterior midgut cell size, as compared to controls.
Adulthood-only expression under the combined control of Scer\GAL4tub.PU and Gal80[ts] leads to the failure to induce autophagy (assessed by the lysosome and autophagosome marker, Lysotracker) in the adult fat body in response to starvation, but has no obvious effect on the intestinal barrier function (as shown by the expected non-permeability to a non-absorbable food dye), as compared to controls.
Embryos derived from females expressing Atg7HMS01358 under the simultaneous control of Scer\GAL4otu.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4nos.UTR.T:Hsim\VP16 show a significant delay in the clearance of the paternal mitochondrial derivatives compared to wild type.
