Description

Imprecise excision of P{EP}SIK2G366 results in a 1,148bp deletion within the SIK2 gene, including the translation start site and the ATP-binding site of the kinase domain.

One-week old SIK2Δ41 mutants exhibit higher amounts of lipid and stored glycogen when compared with controls. This difference becomes more apparent after 24 hours of starvation. These flies do not exhibit a difference to controls in body weight or food intake.

Under starvation conditions, the median and maximum survival times increased by 26 and 20% respectively, in SIK2Δ41 flies when compared to controls.

A TORC25-3 heterozygous background decreases the lipid and glycogen levels in a fed and starved SIK2Δ41 mutant. A TORC25-3 homozygous background further decreases the lipid and glycogen levels in a fed and starved SIK2Δ41 mutant. As the level of TORC decreases, the flies become more sensitive to starvation.

Expression of TORCS157D.Scer\UAS.T:Zzzz\FLAG (under the control of Scer\GAL4hs.PB) in fed and starved SIK2Δ41 mutants reduces the lipid and glycogen levels.

As CrebB-17A levels are decreased (through expression of CrebB-17AKK108927 under the control of Scer\GAL4elav-C155) the increased lipid stores and starvation resistance of SIK2Δ41 flies are strongly inhibited.

Neuronal expression of SIK2Scer\UAS.T:Hsap\MYC (under the control of Scer\GAL4elav-C155) in a SIK2Δ41 background reduces the glycogen and lipid content levels to those seen in controls in both fed and starved conditions.

Fat-body expression of SIK2Scer\UAS.T:Hsap\MYC (under the control of Scer\GAL4Cg.PA) in a SIK2Δ41 background does not alter either the glycogen or lipid content levels.

A dramatic decrease in starvation resistance to the level found in wild-type controls is observed in SIK2Δ41 mutants after expression of exogenous SIK2Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav-C155 but not Scer\GAL4Cg.PA.
