P-element excision results in a small deletion leading to removal of the first 148 codons of the Pak3 A isoform.
1296 bp deletion resulting from the imprecise excision of P{EP}Pak3EP1191, extends from the insertion site to 443 bases beyond the ATG.
Pak376A homozygous mutant embryos exhibit various cuticle phenotypes including head holes and defects in the dorsal surface that range from germband retraction failure to dorsal holes and puckers.
Pak376A has embryonic/larval cuticle phenotype, enhanceable by Pak14/Pak14
Pak376A has embryonic/larval cuticle phenotype, enhanceable by Pak[+]/Pak14
Pak376A, Pak14 has embryonic/larval cuticle phenotype, suppressible | partially by Pak3UAS.GFP/Scer\GAL4hs.PU
Pak3[+]/Pak376A is an enhancer of embryonic/larval cuticle phenotype of Pak14
Pak376A, Pak14 has embryonic leading edge cell phenotype
Pak376A, Pak14 has embryonic dorsal epidermis phenotype
Pak376A, Pak14 has adherens junction phenotype
Pak376A, Pak14 has septate junction phenotype
Pak14 enhances the cuticle phenotypes seen in Pak376A homozygous embryos, with nearly all Pak14 Pak376A double mutants exhibiting head holes. A similar but less severe phenotype is seen in Pak376A homozygous mutants with one copy of Pak14 and Pak14 homozygous mutants with one copy of Pak376A.
Expression of PakScer\UAS.T:Hsap\MYC under the control of Scer\GAL4hs.PU at 29[o]C rescues the defects in the dorsal surface of the embryonic cuticle seen in Pak14 Pak376A double mutants, but does not effectively suppress the head hole phenotype.
The dorsal vessel in Pak376A Pak14 double mutant embryos develops normally.
In Pak376A Pak14 double mutants the opposing leading edge membranes come together in a zig zag pattern following dorsal closure, giving the dorsal midline a cleft-like appearance. In a small number of embryos the epidermal flanks appear to come apart following dorsal closure.
Unlike in wild type, neither septate or adherens junctions are restored at the leading edge following dorsal closure in Pak376A Pak14 double mutants.
