Description

4161bp deletion within prd1 that deletes most of the coding region (amino acids 329-1354).

CG17360M56 is an intragenic deletion generated from PBac{WH}CG17360f05458 and PBac{RB}CG17360e02295. The associated recombinant transposon insertion is PBac{RB3.WH3}CG17360M56.

Comment:

Scer\FRT-mediated recombination between the two progenitor insertions has resulted in the deletion of the genomic sequence between them.

prd1M56/Df(3R)Exel7310 and prd1M56/Df(3R)PS2 transheterozygotes exhibit severe and highly penetrant ddaC neuron dendrite pruning defects at 16h after puparium formation, as compared to wild-type and prd1M56 heterozygous controls.

prd1M56/Df(3R)Exel7310 transheterozygous white prepupal ddaC neurons do not show dendritic arborization defects, whereas prd1M56 ddaC neuron clones in white prepupae show a simplified dendritic tree (i.e. a significant decrease in dendritic termini), as compared to controls.

prd1M56 ddaD/E neuron clones within mosaic individuals also exhibit severe dendrite pruning defects at 16h after puparium formation. ddaF neuron clones, however, are eliminated by 16h after puparium formation, similarly to controls.

The pruning defects observed in prd1M56/Df(3R)Exel7310 transheterozygous ddaC neurons at 16h after puparium formation are significantly enhanced by heterozygosity for unc-104170, unc-104172 or AP-2α3, as the persisting dendrites become significantly longer.

Likewise, the ddaC neuron pruning defects induced by the Scer\GAL4ppk.PU-driven expression of AP-2αHMS00653 are significantly enhanced by heterozygosity for prd1M56, as the persisting dendrites become significantly longer, but are not affected by heterozygosity for Mical15256 or Cul1EX.
