Terminal tracheal cell clones mutant for ich206 in mosaic third instar larvae display seamless tube defects, namely discontinuous membrane-bounded lumens with an irregular cystic appearance. Scer\GAL4btl.PS-driven expression of ichScer\UAS.cRa in the mosaic larvae strongly rescues the tube discontinuity in the mutant cells but gain-of-function morphological defects such as terminal cell pruning are also observed.

ich206 mutant embryos exhibit mild 'blimp' (bloated) defects in cuticular preparations along with reduced sclerotization of the head skeleton and ventral epidermal denticles.

ichor206 mutants exhibit a terminal cell gas-filing defect. They are defective in matrix clearance in all seamless tubes: mutant terminal cels have normal morphology and form seamless tubes, but the tubes fail to clear and gas-fill.

The integrity defects of the seamless tube in terminal tracheal cell clones mutant for ich206 in third instar larvae is weakly suppressed by Scer\GAL4btl.PS-driven expression of lintScer\UAS.cRa - about 2% of clones are rescued and that only if the larvae are kept at 29[o]C to boost the transgene expression.

Comments

The discontinuous seamless tube phenotype observed in terminal tracheal cell clones mutant for ich206 in mosaic third instar larvae is partially rescued by Scer\GAL4btl.PS-driven expression of any of the following ichScer\UAS.cRa, ichDBD.Scer\UAS.T:Zzzz\FLAG.T:SV40\nls-largeT.T:Hsim\VP16 or ichScer\UAS.T:Zzzz\FLAG : the proportion of ich206 terminal cells with seamless tube discontinuities is much decreased.
