wkd220/Df(3R)Exel6276 mutants exhibit overly exuberant tracheal seamless tube growth. Examination of wkd220/Df(3R)Exel6276 terminal cell tips reveals a "U-turn" phenotype in which seamless tubes execute a series of 180[o] turns. All other aspects of tracheal development appear normal at the third instar larval stage. wkd220 mutant clones in both multicellular and unicellular tubes are of normal morphology. wkd220 mutant fusion cells also appear normal.

wkd220 mutants exhibit variable terminal cell pruning and defects in lumen formation, including prematurely truncated tubes that end in local dilations and discontinuous tubes.

Expression of Rab35S22N.Scer\UAS.T:Avic\GFP-YFP under the control of Scer\GAL4btl.PS strongly rescues the lethality and "U-turn" phenotype seen in wkd220/Df(3R)Exel6276 mutant terminal tracheal cells.

wkd220/Df(3R)Exel6276 alters the location of the ectopic seamless tube phenotype seen in tracheal terminal cells expressing btlScer\UAS.T:λ\cI-DD under the control of Scer\GAL4btl.PS. In some cells the extra tubes are found throughout the cell (in the soma and at the branch tip), whereas in others extra tubes are present only at the branch tip. A wkd220/+ background has no effect on seamless tube localisation.

wkd220/Df(3R)Exel6276 does not enhance the ectopic seamless tube phenotype seen in Dlic1 mutant tracheal terminal cell clones.

wkd220/Df(3R)Exel6276 does not enhance the ectopic seamless tube phenotype seen in Dlic2 mutant tracheal terminal cells.

Expression of Rab35S22N.Scer\UAS.T:Avic\GFP-YFP under the control of Scer\GAL4btl.PS strongly rescues the "U-turn" phenotype seen in wkd220/Df(3R)Exel6276 mutant terminal tracheal cells. The pupal stage lethality seen in wkd220/Df(3R)Exel6276 mutants is also rescued.

Expression of wkd+t3.9 rescues the tracheal defects and lethality seen in wkd220/Df(3R)Exel6276 mutants.

Expression of wkdScer\UAS.T:Zzzz\eqFP578-mKate2 under the control of Scer\GAL4btl.PS rescues the tracheal terminal cell defects seen in wkd220/Df(3R)Exel6276 mutants.
