Ectopic expression of Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL and in the absence of any tissue damage inhibits eye disc growth in third instar larvae and causes delay in pupariation compared to age-matched controls.

Expression of Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL or Scer\GAL4elav.PU results in delay to pupariation, as compared to controls.

Overexpression of Ilp8Scer\UAS.T:Zzzz\FLAG driven by Scer\GAL4tub.PU delays pupariation, though larvae reach the correct pupal and adult size; when mutant larvae are fed methoprene, pupae significantly larger than controls are produced, pupariation is delayed and lethality occurs.

Scer\GAL4αTub84B.PL-mediated expression of Ilp8Scer\UAS.T:Zzzz\FLAG delays pupariation by 55.9 +/- 7.6 hours.

Although Scer\GAL4αTub84B.PL, Ilp8Scer\UAS.T:Zzzz\FLAG larvae prolong their feeding (longer than controls), they are no larger (by volume) than controls at the time of pupariation. However, their extended feeding makes the adults weigh more than controls.

The eye disc growth inhibition as well as the pupariation delay (in the absence of tissue damage) observed in third instar larvae expressing Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL are suppressed by co-expression of Lgr3GL01056 (together with UAS-Dicer2 to enhance RNAi efficiency).

Expression of Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4arm.PS in a Lgr3ag1/+ background results in delay to pupariation, as compared to controls, but this delay is partially suppressed in a Lgr3ag1/Lgr3ag1, Lgr3ag1/Lgr3ag8, Lgr3ag1/Lgr3ag9 or Lgr3ag1/Df(3R)BSC321 background.

Co-expression of Lgr3JF03217 partially suppresses and Lgr3GL01056 fully suppresses, but co-expression of both Lgr3GL01056 and Scer\GAL80elav.PU fails to suppress, the developmental delay seen in flies expressing Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL.
