Nucleotide change:

Amino acid change:

K303term | beag-PA

Reported amino acid change:

?304

Comment:

Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change. The mutation was mapped to codon 303. A single point mutation at codon 304 would not result in a nonsense mutation.

beag1 mutants are adult semi-lethal with 39% of the expected number surviving to adulthood.

beag1 and beag1/Df(3R)Exel6151 larvae have decreased numbers of NMJ synaptic boutons compared to wild type, but a significant increase in the area of boutons present, with the result that the total presynaptic area is increased compared to wild type.

beag1/beagEP3260 larvae have decreased numbers of NMJ synaptic boutons compared to wild type.

beag1/+ NMJs have no change in bouton number compared to controls.

Only 6.7% if beag1 NMJs have new boutons added to their distal ends after 48 hours, compared to 51.4% in wild type.

beag1/beagEP3260 animals have a 47% decrease in EJP amplitude at the larval NMJ compared with wild type. (This difference is exaggerated at high calcium concentrations.) mEJP amplitude and frequency are unchanged, leading to a 34% decrease in quantal content compared with wild type.

beag1/Df(3R)Exel6151 NMJ terminals have no difference in the number of brp-positive active zones compared with wild type.

Animals in which beag1 is heterozygous and the Smu1 locus is hemizygous (using Df(3R)Exel6182) show an 11% decrease in bouton number compared with either single mutant.

Scer\GAL4elav-C155-mediated overexpression of Smu1Scer\UAS.cBa does not alter bouton number of beag1/Df(3R)Exel6151 mutants.

Animals heterozygous for both Fas2e76 and beag1 have a 42% decrease in NMJ synaptic bouton number compared with wild type, whereas the single heterozygotes show no change in bouton number. Combining beag1/Df(3R)Exel6151 with heterozygous or homozygous Fas2e76 does not further decrease bouton number compared with beag1/+ alone.

Scer\GAL4Rapgap1-OK6-mediated expression of Fas2Scer\UAS.cLb, but not Fas2Scer\UAS.cLa or Fas2C.Scer\UAS, rescues synaptic bouton number and average synaptic bouton area of beag1/Df(3R)Exel6151 animals to wild type.

Scer\GAL4Rapgap1-OK6-mediated expression of Fas2Scer\UAS.T:Hsap\CD8A rescues synaptic bouton number of beag1/Df(3R)Exel6151 animals to wild type.

Scer\GAL4Rapgap1-OK6-mediated expression of Fas2PEST+.AAE.Scer\UAS rescues synaptic bouton number of beag1/Df(3R)Exel6151 animals to wild type.

Comments

beagT:Zzzz\FLAG rescues beag1/Df(3R)Exel6151 synapse morphology defects and viability.

Expression of beagScer\UAS.cBa using Scer\GAL4Rapgap1-OK6 or Scer\GAL4elav-C155, but not Scer\GAL4G14, fully rescues the decreased bouton number observed in beag1/Df(3R)Exel6151 animals.

Expression of beagScer\UAS.T:Avic\GFP-EYFP using Scer\GAL4elav-C155 fully rescues the decreased bouton number observed in in beag1/Df(3R)Exel6151 animals.

Expression of beagScer\UAS.cBa using Scer\GAL4elav-C155 or Scer\GAL4VGlut-OK371 rescues beag1 adult viability.

Expression of beagScer\UAS.cBa using Scer\GAL4Rapgap1-OK6 fully rescues the reduction in EJP amplitude and quantal content in beag1/beagEP3260 mutants.
