ppk23Δ8.284 mutant males show significantly lower courtship intensity as compared with wild type.

Homozygous ppk23Δ8.284 mutant larvae have significantly reduced preference for larval-treated substrate, compared to controls; attraction to the secreted larval fatty acids (Z)-5-tetradecanoic acid and (Z)-7-tetradecanoic acid is significantly reduced, compared to controls.

ppk23Δ8.284 males show vigorous courtship towards wild-type target males, as measured by the number of unilateral wing extensions. Moreover, when 6-9 ppk23Δ8.284 males are placed together in a chamber, they serially court each other, forming long chains in which males follow each other and produce courtship songs.

ppk23Δ8.284 males lacking antennae show a complete loss of male-male courting.

In a 30min trial, ppk23Δ8.284 males court females with reduced frequency and longer latency compared to controls, often with ppk23Δ8.284 males failing to court at all.

ppk23Δ8.284 mutant flies explosed to oe[-] males (Scer\GAL4desat1.PB, Scer\GAL80ts.αTub84B/WScer\UAS.cZa) painted with 7T, 7P, or cVA, do not exhibit differences in courtship, whereas control males exhibit a reduction in courtship. Additionally, controls increase courtship to oe[-] females painted with 7,11-HD, 7,11-ND, or 7P, whereas ppk23Δ8.284 males show no change in behavior.

Introduction of ppk29Scer\UAS.cTa into ppk23Δ8.284 mutants under the control of Scer\GAL4ppk23.2.695 fails to suppress the courtship defects found in ppk23Δ8.284 mutants.

In proboscis extension assays, ppk23Δ8.284 ppk29Δ double mutant flies show no significant defects in detection of sugars, bitter compounds, salt, or water.
