UAS regulatory sequences drive expression of the full-length Dana\Khc coding sequence. Two stretches of perfect homology (21 and 17 nucleotides long) that is targeted by an RNAi construct against Khc (KhcGD12278) have been changed by site-directed mutagenesis to improve rescue ability.
Dana\KhcUAS.RR/Scer\GAL4da.G32 is a suppressor of lethal phenotype of Khc8/Df(2R)Jp6
Dana\KhcUAS.RR, Scer\GAL4repo.PL, Scer\GAL4repo is a suppressor of abnormal locomotor behavior | adult stage phenotype of KhcGD12278, Scer\GAL4repo.PL, Scer\GAL4repo
Dana\KhcUAS.RR, Scer\GAL4repo.PL, Scer\GAL4repo is a suppressor of abnormal locomotor behavior | larval stage phenotype of KhcGD12278, Scer\GAL4repo.PL, Scer\GAL4repo
Dana\KhcUAS.RR, Scer\GAL4Gli-J29-Gal4 is a suppressor of abnormal neurophysiology | third instar larval stage phenotype of KhcGD12278, Scer\GAL4Gli-J29-Gal4
Co-expression of Dana\KhcRNAi-res.Scer\UAS rescues the locomotor defects seen in adults and larvae expressing KhcGD12278 under the simultaneous control of both Scer\GAL4repo.PL and Scer\GAL4repo.
Expression of Dana\KhcRNAi-res.Scer\UAS under the control of Scer\GAL4da.G32 rescues the lethality of Khc8/Df(2R)Jp6 flies.
Co-expression of Dana\KhcRNAi-res.Scer\UAS rescues the reduced excitatory junction potential amplitude seen in third instar larvae expressing KhcGD12278 under the control of Scer\GAL4Gli-J29-Gal4.
