Description

The endogenous stck promoter drives expression of a mutant form of stck in which the glutamine at amino acid 38 has been replaced by alanine. The construct is tagged at the C-terminal end with Tag:FLAG.

Expression of stckQ38A.T:Zzzz\FLAG in an otherwise wild-type background has no effect on larval size.

No obvious phenotypes are seen when stckQ38A.T:Zzzz\FLAG is expressed in wild type flies.

Embryos expressing stckQ38A.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background do not display any muscle structure or actin cytoskeleton defects.<D-c>

flwG0172 mutants carrying stckQ38A.T:Zzzz\FLAG arrest during the larval stages. The larvae are significantly smaller and significantly less motile than flwG0172 single mutant larvae. Expression of stckQ38A.T:Zzzz\FLAG does not rescue the detachment of ventral intersegmental muscles seen in flwG0172 larvae.

The addition of icsBG02577 reduces the viability of flies expressing stckQ38A.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background. Between 6 and 34% of the expected number of progeny are seen, with the majority of the lethality taking place during the larval and pharate stages. The rescued larvae display a range of sizes, including presumptive first instar larvae that appear stumpy, exhibit sluggish movement and do not progress in development.
