Description

Excision of the FRT-w cassette from progenitor AxnH504L.FRT.αTub84B.Tag:FLAG allows the αTub84B promoter to drive expression of an H504L mutant of Axn tagged with N-terminal 6xTag:FLAG.

More than half of AxnS044230 (maternal-/zygotic-) mutant embryos expressing maternally provided AxnH504L.αTub84B.T:Zzzz\FLAG hatch successfully.

AxnS044230 mutant (maternal-/zygotic-) embryos expressing maternally provided AxnH504L.αTub84B.T:Zzzz\FLAG display normal cuticle patterning.

Zygotic AxnS044230 mutants receiving maternally deposited wild-type Axn and expressing AxnH504L.αTub84B.T:Zzzz\FLAG survive to adulthood without defects. Subsequently, if the rescued animals are mated with each other, they produce a few larvae and misshapen pupae that subsequently die. This indicates that both sexes of the parents are fertile.

AxnH504L.αTub84B.T:Zzzz\FLAG combined with AxnΔRGS.αTub84B.T:Zzzz\FLAG can rescue AxnS044230 mutants to adult viability and normal morphology.

AxnH504L.αTub84B.T:Zzzz\FLAG combined with AxnK446E.αTub84B.T:Zzzz\FLAG can fully rescue AxnS044230.

Expression of maternally provided AxnH504L.αTub84B.T:Zzzz\FLAG decreases the hatching rate of embryos to 81.9%.

Zygotic expression of AxnH504L.αTub84B.T:Zzzz\FLAG does not interfere with development and survival. Adults show no visible defects.
