Utx sequences (residues 503-952) were deleted with ends-out gene targeting and replaced with a mini-white gene and the AGC codon for Ser503 was replaced with an in-frame TAA termination codon after the His502 codon of Utx.

UtxΔ/UtxΔ adults derived from UtxΔ/+ mothers die within 1-2 days post eclosion. When derived from UtxΔ/+ mothers with homozygous UtxΔ female germline clones, UtxΔ/UtxΔ flies die during larval stages.

Adults carrying the Utxg.wt allele in a UtxΔ/UtxΔ background derived from UtxΔ/+ mothers both with or without homozygous UtxΔ female germline clones can be maintained as a healthy strain.

Adults carrying the Utxg.cd allele in a UtxΔ/UtxΔ background derived from UtxΔ/+ mothers appear morphologically indistinguishable from wild-types, but die within 1-2 days post eclosion.

UtxΔ mutants are homozygous lethal. UtxΔ enhances the variegated phenotype of the w locus in the In(1)wm4 background but has no effect on the variegation in the Sb locus when combined with T(2;3)SbV.

The majority of homozygous UtxΔ and UtxΔ/Df(2L)BSC143 animals that are derived from UtxΔ/+ females develop into adults that are morphologically indistinguishable from wild type, but die shortly after eclosion.

Homozygous animals derived from females carrying homozygous female germline clones arrest development before the pupal stage; 85% die before reaching the third larval instar and the remaining animals die during the third larval instar, with only 0.6% developing to form white prepupae, which than also die. This maternal effect lethality can be paternally rescued: animals derived from homozygous oocytes fertilised by wild-type sperm are viable and fertile. The majority of the rescued adults have normal morphology, but 30% of males show a patchy loss of pigmentation in the tergite of the fifth abdominal segment (representing a transformation into a more anterior segment) and 2.8% of individuals show transformation of parts of the haltere into wing tissue.

Homozygous clones in the eye-antennal imaginal disc do not show an overproliferation phenotype.
