Expression of Hdac3dsRNA.Sym.Scer\UAS under the control of Scer\GAL4ey.PH results in a reduction in the size of the eye. The average area of male fly eyes is reduced by approximately 30%, compared to controls. The depletion of Hdac3 results in an 11% reduction in ommatidial size. The total number of ommatidia is also reduced, with an approximately 24% reduction.

Expression of Hdac3dsRNA.Sym.Scer\UAS under the control of Scer\GAL4e22c results in a reduction in the posterior compartment of the wing.

Expression of Hdac3dsRNA.Sym.Scer\UAS under the control of Scer\GAL4ppl.PP in the fat body results in progeny that are partially lethal, with the surviving progeny exhibiting a small body size at the 3rd-instar larval, pupal and adult stages. The average weight of the Hdac3 RNAi female progeny is ~16% less than that of controls, while the weight of male progeny is reduced by ~14%.

Depletion of Hdac3 in the eye, through expression of Hdac3dsRNA.Sym.Scer\UAS under the control of Scer\GAL4GMR.PF, strongly suppresses the overgrown, disorganised eye phenotype (with increased ommatidial size) found upon expression of InRScer\UAS.Exel in the eye. These double-mutant flies exhibit a phenotype similar to Hdac3dsRNA.Sym.Scer\UAS single mutants.

Overexpression of His4K16A.Scer\UAS in a Hdac3dsRNA.Sym.Scer\UAS background partially attenuates the Hdac3dsRNA.Sym.Scer\UAS-induced decrease in eye size. Consistent with this finding, the reduction in body weight induced by Hdac3dsRNA.Sym.Scer\UAS is also partially attenuated by co-expression of His4K16A.Scer\UAS.
