Expression of Itp-r83ANIG.1063R under the control of Scer\GAL4elav-C155 leads to significant decrease in flight ability in adults (measured as flight time in a single flight assay on tethered flies).

A partial flight deficit is observed upon expression of Itp-r83ANIG.1063R in dopaminergic neurons under the control of Scer\GAL4ple.PF. The flight deficits correlated with abnormalities in action potentials recorded from dorsal longitudinal muscles during air puff-induced flight bouts.

Pupal expression of Itp-r83ANIG.1063R in all dopaminergic neurons leads to partial flight deficits.

Flies expressing Itp-r83ANIG.1063R under the control of Scer\GAL4Trh.PB show normal flight ability in a cylinder drop test. However, the flies show increased spontaneous firing from the dorsal longitudinal indirect flight muscles compared to wild type.

Pan-neuronal expression of Itp-r83ANIG.1063R driven by Scer\GAL4elav-C155 results in partial pupal lethality. The surviving Scer\GAL4elav-C155; Itp-r83ANIG.1063R flies exhibit wing posture defects. Scer\GAL4elav-C155; Itp-r83ANIG.1063R mutant flies show significant flight defects and are unable to initiate rhythmic flight patterns in recordings from the dorsal longitudinal muscles in response to air-puff stimulus.

No significant change in wing posture or flight ability is observed when Itp-r83ANIG.1063R is expressed with either Scer\GAL4Ilp2.PR or Scer\GAL4Ddc.PL.

Spontaneous firing is increased from the dorsal longitudinal muscles when Itp-r83ANIG.1063R is expressed with either Scer\GAL4elav-C155 or Scer\GAL4Ilp2.PR or Scer\GAL4Ddc.PL.

In primary neuronal cultures obtained from brains of larvae expressing Itp-r83ANIG.1063R driven by Scer\GAL4elav-C155, store operated Ca2[2+] entry is reduced compared to controls.

Store Ca[2+] or "["Ca[2+]"]"[[ER]] is similar in primary neuronal cultures between brains from larvae expressing Itp-r83ANIG.1063R driven by Scer\GAL4elav-C155 and controls.

Expression of Itp-r83ANIG.1063R under the control of Scer\GAL4Act5C.PI results in significantly smaller larvae compared to controls at 120 hours after egg laying. At an earlier stage, 85 hours after egg laying, mutant larvae are similar in size to control larvae.

Expression of Itp-r83ANIG.1063R under the control of Scer\GAL4elav-C155, Scer\GAL4Ddc.PL or Scer\GAL4Ilp2.PR (and in the presence of Dcr-2Scer\UAS.cDa) does not affect larval size, and mutant larvae are similar in size to control larvae, and the number of pupae formed is similar in number to controls.

Expression of Itp-r83AdsRNA.Scer\UAS in the mushroom bodies, under the control of Scer\GAL4OK107 results in defects in the mushroom bodies in approximately 24% of cases (compared to 13% when only Itp-r83AdsRNA.Scer\UAS is present) and defects in the antennal lobes in 5% of cases. No defects were observed in the optic lobes.

Flies expressing Itp-r83ANIG.1063R and Dcr-2Scer\UAS.cDa driven by both Scer\GAL4Ilp2.PR and Scer\GAL4Ddc.PL show normal wing posture, but they are unable to maintain flight beyond 15-20 s.
