UAS regulatory sequences are fused upstream of sequence encoding the FKBP12 destabilization domain (Tag:degron(FKBP)) followed by GAL4::VP16 driver sequence. This construct was intended to result in a GAL4::VP16 driver whose degradation could be controlled chemically by the 'Shield1' small molecule (which can bind the Tag:degron(FKBP) sequence to prevent degradation, see PMID:16959577), but the destabilization domain does not appear to be effective in the P{UAS-DD-GAL4.VP16.I}CG60511 insertion of this construct. The P{UAS-DD-GAL4.VP16.I}CG60511 insertion also does not behave as expected with respect to UAS-driven expression, and it is not known whether other insertions of this construct will show the expected UAS-inducible expression.
ScerGAL4VP16.UAS.cIa expresses GAL4 in the late pupal wing blade, and in larval muscles and trachea (presumably reflecting nearby enhancers), but no apparent UAS activity. Destabilization domain appears ineffective.
