UAS regulatory sequences drive expression of a full-length Tgs1 open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.

Expression of Tgs1Scer\UAS.ORF.GW.T:Ivir\HA1 under the control of any of the following: Scer\GAL4Mef2.PR, Scer\GAL4how-24B, Scer\GAL4da.PU and Scer\GAL4c179 leads to complete lethality or reduced adult viability at 25[o]C and 29[o]C with more severe effects being observed at the higher temperature. Expression driven by Scer\GAL4G7 or Scer\GAL4elav.PU or Scer\GAL4RapGAP1-OK6 does not significantly decrease adult viability at either of the two temperatures.
Expression of Tgs1Scer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4how-24B results in significant decrease in the rate of body wall muscle contractions in third instar larvae and increased length (length/width axial ratio) of pupae compared to controls.
Co-expression of Tgs1Scer\UAS.ORF.GW.T:Ivir\HA1 rescues the lethality of flies expressing Tgs1GD14932 under the control of Scer\GAL41032.hx.
