Nlg3 genomic region (nucleotides -1556 to +2293, boundaries inferred from primer sequences) have been replaced by a P1\loxP cassette containing a w[+mW.hs] marker.
Excitatory junctional potential (EJP) amplitude and miniature excitatory junctional potential (mEJP) frequency are significantly reduced at the Nlg3KO88/Nlg3f00777 third larval instar neuromuscular junction compared to wild type. mEJP amplitude is not significantly affected.
Nlg3KO88/Nlg3f00777 third larval instar larvae show a significant increase in bouton number and a significant decrease in bouton size at the neuromuscular junction compared to wild type. Synaptic branch number at the neuromuscular junction is not significantly affected.
Ultrastructurally, the neuromuscular junction of Nlg3KO88/Nlg3f00777 larvae shows a number of defects compared to wild type. There is a shift in the distribution of synaptic vesicle diameter to larger values and the number of large vesicles greater than 80nm in diameter is dramatically increased. The number of vesicles at the active zone is significantly decreased, and the postsynaptic density length is significantly increased.
Synaptic endocytosis at the neuromuscular junction is impaired in Nlg3KO88/Nlg3f00777 larvae.
Nlg3KO88/Nlg3KO127 is rescued by Nlg3UAS.cXa/Scer\GAL4Mhc.PU
Muscle-specific expression of Nlg3Scer\UAS.cXa (under the control of Scer\GAL4Mhc.PU) rescues the defects in bouton number and size seen at the neuromuscular junction in Nlg3KO88/Nlg3KO127 larvae. Neuron-specific expression (under the control of Scer\GAL4elav.PU) does not rescue this phenotype.
