Description

Amino acid replacement: R245A.

Amino acid replacement (R245A) in the O-fut1 GDP-fucose binding site. A single loxP site is also present due to the method used to generate the allele.

O-fut1R245AKI homozygous third instar larvae exhibit a strong increase in the number of wing disc sensory organ precursor cells at the non-permissive 30[o]C of temperature, but not at the permissive 25[o]C of temperature, associated to a significant increase in number of supraalar neurons, which is bigger at 30[o]C than at 25[o]C, as compared to controls.

O-fut1R245AKI homozygotes survive until the third-instar larval stage.

O-fut1R245AKI homozygotes obtained from females carrying the O-fut1R245AKI homozygous germline show a temperature-sensitive neurogenic phenotype. At 30[o]C, O-fut1R245AKI m/z embryos show a highly penetrant neurogenic phenotype.

O-fut1R245AKI homozygous wing disc clones exhibit an increase in the number of sensory organ precursors at 30[o]C, but display wild-type numbers at 18 and 25[o]C.

The third instar larval wing discs of O-fut1R245AKI/O-fut1R245AKI, Uxs1/+ individuals or O-fut1R245AKI/O-fut1R245AKI, shamsΔ34/shamsΔ34 individuals, but not O-fut1R245AKI/O-fut1R245AKI, Uxs1/Uxs1 individuals, raised at 25[o]C of temperature, exhibit increased numbers of both sensory organ precursor cells and supraalar neurons, as compared to controls.

The increased number of supraalar neurons present in the third instar larval wing discs of O-fut1R245AKI homozygotes raised at 25[o]C of temperature is strongly enhanced by rumi44 homozygosity.

O-fut1R245AKI and rumi44 homozygous double mutant wing disc clones exhibit an increase in the number of sensory organ precursors at 18, 25 and 30[o]C.
