Expression of Diap1dsRNA.UAS.cLa under the control of Scer\GAL4GMR.PFa results in significant eye size reduction and depigmentation.

Male flies expressing thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 exhibit slightly increased levels of spermatogonial cyst death compared to controls.

Scer\GAL4GMR.PF-mediated expression of thdsRNA.Scer\UAS.cLa induces massive degeneration reflected in disrupted pigmentation and reduction and roughening of the eye.

Approximately 5% of Scer\GAL4GMR.PF,thdsRNA.Scer\UAS.cLa animals die as pupae.

Compared with wild-type, third instar larvae expressing thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4ap-md544 display signs of increased apoptosis in the dorsal compartment of wing imaginal discs.

Expression of thdsRNA.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF results in reduced, deformed and partially depigmented eyes.

Expression of thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4GMR.PF results in a small eye phenotype.

Expression of thdsRNA.Scer\UAS.cLb in the eye under the control of Scer\GAL4GMR.PF results in massive cell death in third instar larval eye discs and severely deformed adult eyes. Adult eyes show a spotted-eye phenotype, distorted outer eye morphology, and no defined interior eye structures develop. Death of eye pigment cells occurs, and loss of eye bristles.

The eye phenotype (depigmentation and size reduction) characteristic for flies expressing Diap1dsRNA.UAS.cLa under the control of Scer\GAL4GMR.PFa is slightly improved by co-expression of Usp15-31KK101035 or by combination with a single copy of Usp15-31KG04149 and is strongly suppressed by co-expression of DarkKK104215.

Scer\GAL4GMR.PF-mediated expression of thdsRNA.Scer\UAS.cLa results in a more severe eye phenotype and increased pupal lethality in a homozygous wunspecified background compared to a wunspecified/+ background.

Expression of P{Sym-UAS-Hsrω} leads to recovery of eye size and morphology in Scer\GAL4GMR.PF,thdsRNA.Scer\UAS.cLa flies, though the ommatidial lattice is still disrupted. Pupal lethality is also suppressed.

Co-expression of HsrωEP93D or HsrωEP3037 leads to a slight enhancement of the eye phenotype in Scer\GAL4GMR.PF,thdsRNA.Scer\UAS.cLa flies. Co-expression of HsrωEP93D suppresses, while co-expression of HsrωEP3037 has no effect on the pupal lethality of Scer\GAL4GMR.PF,thdsRNA.Scer\UAS.cLa flies.

Expression of thdsRNA.Scer\UAS.cLa using Scer\GAL4GMR.PF enhances the rprGMR.PW and grimGMR.PH small eye phenotypes.

Co-expression of thdsRNA.Scer\UAS.cLa enhances the eye phenotype of Scer\GAL4GMR.PF, egrScer\UAS.cIa flies.

Co-expression of Hsp60DdsRNA.Sym.Scer\UAS has no effect on the eye phenotypes caused by expression of thdsRNA.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF.

Co-expression of Hsp60DScer\UAS.cAa mildly enhances the eye phenotypes caused by expression of thdsRNA.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF.

The small eye phenotype observed in animals expressing thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4GMR.PF is suppressed by co-expression of Iap2Scer\UAS.cRa, Iap2C46G.Scer\UAS, or Iap2C149G.Scer\UAS. Co-expression of Iap2C249G.Scer\UAS or Iap2C466Y.Scer\UAS fails to suppress this phenotype.

The death of eye pigment cells observed in animals expressing thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4GMR.PF is enhanced in a Df(3L)H99/Df(3L)H99 mutant background.

Comments

The eye phenotype in animals expressing thdsRNA.Scer\UAS.cLb under the control of Scer\GAL4GMR.PF is fully rescued by co-expression of thScer\UAS.cHa.
