Heterozygous esgL4 mutant flies exhibit increased sensitivity to nicotine. Approximately 8% of heterozygous esgL4 flies exhibit cuticle defects such as loss of bristles in the tergites and sternites, and loss of pigmentation in the dorsal abdomen. Responses to heat and ether are normal.
Expression of esgHMS00025 under the control of Scer\GAL4L4 enhances the cuticle phenotypes seen in heterozygous esgL4 mutant flies from 8% to 100%. The resulting flies are not viable.

Expression of mir-311Scer\UAS.T:Ppyr\LUC under the control of Scer\GAL4L4 enhances the cuticle phenotype seen in esgL4/+ mutant flies, with 23% of the resulting flies having phenotypes compared with 8% in esgL4/+ alone. Viability is unaffected. The increase in nicotine sensitivity is also enhanced.
Expression of mir-14Scer\UAS.T:Ppyr\LUC under the control of Scer\GAL4L4 suppresses the cuticle phenotype and nicotine sensitivity seen in esgL4 mutant flies. Viability is unaffected.
One copy each of P{GawB}mir-310L70 (comprised of mir-310L70, mir-311L70 and mir-312L70) and esgL4 results in increased sensitivity to nicotine compared to esgL4/+ alone.
