Tudor-SN1120/Df(3L)BSC125 (2-day old males) have swollen apical tips of testes containing excessive numbers of early spermatogenic cells (86% show this phenotype). Tudor-SN0614/Tudor-SN1120 (81%) or Tudor-SN1120/Tudor-SN1236 (78%) transheterozygotes show the same phenotype. Ovaries appear normal in Tudor-SN0614/Tudor-SN1120 females.

Tudor-SN0614/Tudor-SN1120 show an over-proliferation of cells (likely spermatocytes) in the swollen apical tip of testes; a few bundles of mislocalized elongated spermatids with heads residing at the apical tip of the testis are also present (62% display this phenotype). Germline stem cell number is similar to wild type. Mutant third instar larval testes have a higher mitotic index of spermatogonia and are larger than wild type larval testes. In Tudor-SN0614/Tudor-SN1120 mutant testes, round onion stage spermatids containing a single over-sized Nebenkern associated with two haploid nuclei are observed; 2 week old mutant adult testes have fewer spermatid bundles compared to wild type. Overall, mutant testes have higher proliferation of spermatogonia but compromised differentiation of spermatocytes.

piwi1/+ (or even more so, piwi1/piwi2) significantly partially suppresses the swollen apical tip of the testis phenotype seen in Tudor-SN0614/Tudor-SN1120 males. piwi1/piwi2 suppresses infertility seen in Tudor-SN0614/Tudor-SN1120 males. Tudor-SN0614/Tudor-SN1120 partially suppresses ovary defects in piwi1/piwi2 females. Expression of piwiGD11827 driven in the germline by Scer\GAL4nos.UTR.T:Hsim\VP16 (but not Scer\GAL4tj-NP1624) suppresses the testis swollen apical tip phenotype seen in Tudor-SN0614/Tudor-SN1120 males.
