Deletion of nucleotides +3009 to +3067 (coordinates according to CG7008 annotation), which results in a frameshift from amino acid residue 437 onwards.
Nebenkern (with Tudor-SN0614)
spermatid | adult stage (with Tudor-SN0614)
testis (with Df(3L)BSC125)
testis (with Tudor-SN0614)
testis (with Tudor-SN1236)
Tudor-SN1120/Df(3L)BSC125 (2-day old males) have swollen apical tips of testes containing excessive numbers of early spermatogenic cells (86% show this phenotype). Tudor-SN0614/Tudor-SN1120 (81%) or Tudor-SN1120/Tudor-SN1236 (78%) transheterozygotes show the same phenotype. Ovaries appear normal in Tudor-SN0614/Tudor-SN1120 females.
Tudor-SN0614/Tudor-SN1120 show an over-proliferation of cells (likely spermatocytes) in the swollen apical tip of testes; a few bundles of mislocalized elongated spermatids with heads residing at the apical tip of the testis are also present (62% display this phenotype). Germline stem cell number is similar to wild type. Mutant third instar larval testes have a higher mitotic index of spermatogonia and are larger than wild type larval testes. In Tudor-SN0614/Tudor-SN1120 mutant testes, round onion stage spermatids containing a single over-sized Nebenkern associated with two haploid nuclei are observed; 2 week old mutant adult testes have fewer spermatid bundles compared to wild type. Overall, mutant testes have higher proliferation of spermatogonia but compromised differentiation of spermatocytes.
Tudor-SN0614/Tudor-SN1120 has male semi-fertile | progressive phenotype, suppressible by piwi2/piwi1
Tudor-SN0614/Tudor-SN1120 has testis phenotype, suppressible | partially by piwi[+]/piwi1
Tudor-SN0614/Tudor-SN1120 has testis phenotype, suppressible | partially by piwi2/piwi1
Tudor-SN0614/Tudor-SN1120 has testis phenotype, suppressible by Scer\GAL4VP16.nanos.UTR/piwiGD11827
Tudor-SN0614/Tudor-SN1120 has testis phenotype, non-suppressible by piwiGD11827/Scer\GAL4NP1624
Tudor-SN0614/Tudor-SN1120 is a suppressor | partially of ovary phenotype of piwi2/piwi1
piwi1/+ (or even more so, piwi1/piwi2) significantly partially suppresses the swollen apical tip of the testis phenotype seen in Tudor-SN0614/Tudor-SN1120 males. piwi1/piwi2 suppresses infertility seen in Tudor-SN0614/Tudor-SN1120 males. Tudor-SN0614/Tudor-SN1120 partially suppresses ovary defects in piwi1/piwi2 females. Expression of piwiGD11827 driven in the germline by Scer\GAL4nos.UTR.T:Hsim\VP16 (but not Scer\GAL4tj-NP1624) suppresses the testis swollen apical tip phenotype seen in Tudor-SN0614/Tudor-SN1120 males.
Tudor-SN0614/Tudor-SN1120 is partially rescued by Scer\GAL4VP16.nanos.UTR/Tudor-SNUAS.Tag:FLAG
Expression of Tudor-SNScer\UAS.T:Zzzz\FLAG driven by Scer\GAL4nos.UTR.T:Hsim\VP16 partially rescues the swollen testis apical tip phenotype and reduced fertility of Tudor-SN1120/Tudor-SN0614 males.
