Clbn1Q/Clbn1Q flies are viable, but show small developmental delays and shortened lifespans. Mitotic chromosomes from the brains of Clbn1Q/Clbn1Q third instar larvae treated with radiation show significant increases in the number of breaks per nucleus compared to wild type (radiation sensitive). Clbn1Q/Clbn1Q third instar larvae develop significantly more melanotic masses after irradiation compared to wild type.

Wild type larval wing discs show two phases of cell death in response to irradiation: first signs of cell death are clear after 4h and plateau around 8h, with cell death again increasing from 16-24h; irradiated Clbn1Q/Clbn1Q discs show early induction of cell death (though significantly less than wild type) and then there is only a small increase in cell death from 8-24h. Similar to hid05014, irradiated Clbn1Q/Clbn1Q discs show significantly less apoptosis at 4h compared to wild type, and show a similar second phases of induction at 16-24h but at significantly lower levels.

Clbn1Q/Clbn1Q p535A-1-4/p535A-1-4 double mutants show the same degree of radiation sensitivity (increase in the number of breaks per nucleus in mitotic chromosomes in third instar larval brains compared to wild type) as Clbn1Q/Clbn1Q or p535A-1-4/p535A-1-4 mutants alone. Clbn1Q/Clbn1Q p535A-1-4/p535A-1-4 double mutant third instar larval wing discs appear to have almost a complete block of cell death (apoptosis) in response to irradiation. hid05014 Clbn1Q/Clbn1Q double mutants show similar apoptosis induction in wing discs in response to irradiation as either mutant alone. Clbn1Q/Clbn1Q does not suppress the induction of apoptosis seen in hidGMR.PG third instar eye discs. Clbn1Q/Clbn1Q significantly enhances melanotic tumor formation in tefustg/tefustg third instar larvae (36.3%).
