Embryos with expression of SsedsRNA.147.Scer\UAS.P\T driven by Scer\GAL4mat.αTub67C.T:Hsim\VP16 arrest with a single mitotic spindle and around half of embryos lack an X-chromosome in the mitotic spindle; the majority of embryos from females contain polar bodies with 4 X-cent and 4 X-arm signals, arm cohesion persists in anaphase I and centromeric cohesion persists (96% of oocytes in meiosis II are in metaphase II, 56% of embryos with polar bodies retain centromeric cohesion). Failure of anaphase II results in generation two rather than four daughter nuclei at the completion of meiosis (both with a single X-cent signal).
Co-expression of vtdGL00522 leads to polar bodies containing 8 X-cent and 8 X-arm FISH signals in embryos with expression of SsedsRNA.147.Scer\UAS.P\T driven by Scer\GAL4mat.αTub67C.T:Hsim\VP16 (polar body chromosomes replicate but fail to release arm cohesion).
