Eggs with expression of vtdGL00522 driven by Scer\GAL4mat.αTub67C.T:Hsim\VP16 do not hatch. Embryos progress through syncytial divisions that become progressively aberrant and asynchronous, with suggestions of precocious chromosome segregation; eggs appear to complete meiosis, with presence of polar body chromosomes that lack centromeric cohesion in embryos.

Mature stage 14 oocytes with expression of vtdGL00522 driven by Scer\GAL4nos.UTR.T:Hsim\VP16 are indistinguishable from wild type; stage 9-11 oocytes are similar to wild type in terms of arm cohesion during meiosis.

Expression of vtdGL00522 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not have any effect on the synaptonemal complex assembly and centromere clustering (assessed by number of cid foci) in oocytes.

Expression of vtdGL00522 (together with UAS-Dicer2 to enhance RNAi efficacy) under the control of Scer\GAL4en-e16E to induce chromosome instability in the proliferative third instar larval wing disc tissue results in increased frequency of aneuploid metaphasic cells, a marked increase in autophagy (visualized by autophagy markers Atg8 and LysoTracker), oxidative stress (elevated levels of ROS) and cell death (visualized using [*]Casp3).

The expression of vtdGL00522 under the control of Scer\GAL4en.PU (together with Dicer-2, for efficient RNAi) leads to a significant increase in apoptosis in the posterior compartment of the third instar larval wing disc, which is suppressed by co-expressing either TlGL00474, dlJF02825, CatScer\UAS.cAa, Dsp1JF02732, egrKK103432 or bskGD10555.
