Description

UAS regulatory sequences drive expression of the the C-terminal half (amino acid residues 141-383) of the Hsap\MAPT 0N4R splice variant, which is required for binding to microtubules and contains the part of the proline-rich domain. The protein is tagged at the C-terminal end with Tag:FLAG.

      Expression of Hsap\MAPTCter.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4ptc.PU significantly reduces the size of the L3-L4 intervein region (measured at the wing margin) of the adult wing compared to controls; in the third instar larval wing disc the Hsap\MAPTCter.Scer\UAS.T:Zzzz\FLAG cells display abnormal mitosis with monopolar spindles.

      Expression of Hsap\MAPTCter.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU leads to rough eye phenotype (missing interommatidial bristles) in the adults.

      Co-expression of Hsap\MAPTCter.Scer\UAS.T:Zzzz\FLAG together with Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU results in rough eye phenotype (missing interommatidial bristles, ommatidial fusions) in the adults. The phenotype is the same as when two copies of full-length Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG are used.
