Acute activation (10 minute blue light exposure) of Zzzz\PACαScer\UAS.cSLa expressed under the control of Scer\GAL4elav.PU significantly increases arousal thresholds (the minimal vibration required to arouse a quiescent fly) compared to controls.
Continual activation of Zzzz\PACαScer\UAS.cSLa expressed under the control of Scer\GAL4elav.PU (exposure to blue light throughout the day, but not at night) results in chronically decreased responsiveness to mechanical stimuli during both day and night. However, sleep intensity is only significantly increased at night (after the blue light is turned off).
Expression of Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4Or83b.2.642.T:Hsim\VP22 increases response index to blue light-induced stimulation, indicating overexpression larvae are now more attracted (or less repelled) by blue light. Application of white light also induces a repression of the negative phototactic behavior of transgenic larvae.
Expression of Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4Or83b.2.642.T:Hsim\VP22 does not lead to adaptation of ORNs to prolonged blue light stimulation nor prevent the animals from perceiving an additional odor stimulus.
Transgenic larvae expressing Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4Or13a.PC, Scer\GAL4Or30a.PC, Scer\GAL4Or33b.PC, Scer\GAL4Or35a.PC, Scer\GAL4Or45a.PC, Scer\GAL4Or45b.PC, Scer\GAL4Or59a.PF, Scer\GAL4Or63a.PC, Scer\GAL4Or67b.PC, Scer\GAL4Or74a.PC, Scer\GAL4Or85c.PC, or Scer\GAL4Or94b.PC are less repelled by blue or white light (in orientation assays) than wild-type larvae. These flies also show a stronger negative phototaxis effect with blue light rather than white light (this wasn't seen with Scer\GAL4Or33b.PC and Scer\GAL4Or45a.PC drivers).
Transgenic larvae expressing Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4Or13a.PC, Scer\GAL4Or30a.PC, Scer\GAL4Or33b.PC, Scer\GAL4Or35a.PC, Scer\GAL4Or45a.PC, Scer\GAL4Or45b.PC, Scer\GAL4Or59a.PF, Scer\GAL4Or63a.PC, Scer\GAL4Or67b.PC, Scer\GAL4Or74a.PC, Scer\GAL4Or85c.PC, or Scer\GAL4Or94b.PC avoid octyl acetate, as in wild-type.
Expression of Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4Or33b.PC or Scer\GAL4Or45a.PC results in an aversion response to blue light.
Neuronal expression of Zzzz\PACαScer\UAS.cSLa under the control of Scer\GAL4elav.PU results in adult flies without any obvious behavioral phenotype. Irradiating adult flies with blue light (but not other wavelengths) results in periods of hyperactivity and unusual freezing behavior in flies expressing Zzzz\PACαScer\UAS.cSLa but not in wild-type. In addition, grooming behavior is substantially reduced in these flies in the presence of blue light. This behavior is lost a few seconds after switching the flies back to white light.
Egra\PACαUAS.cSLa, Scer\GAL4Orco.2.642.Hsim\VP22 has abnormal phototaxis phenotype, suppressible by norpAP24
A norpA36 background blocks phototaxis in Zzzz\PACαScer\UAS.cSLa-Scer\GAL4Or83b.2.642.T:Hsim\VP22 mutants.
