The expression of Arf79FT31N.Scer\UAS.cWa under the control of Scer\GAL4ppk.PU leads to dorsal multidendritic ddaC neurons exhibiting a significantly simplified dendritic tree at the white pupa stage and a small but significant delay in dendrite pruning at 16h-18h after puparium formation, as compared to controls.
Clonal expression under the control of Scer\GAL4109(2)80 leads to dorsal multidendritic ddaD/E neurons exhibiting a simplified dendritic tree at the white pupa stage and exhibiting defective dendrite pruning, as shown by the retention of some dendrite branches at 19h after puparium formation, as compared to controls.
Expression under the combined control of Scer\GAL4GSG2295 and RU486 treatment from mid-third instar larval stage onwards leads to dorsal multidendritic ddaC neurons exhibiting a fully penetrant pruning defect, as shown by the persistence some dendritic branches at 16h-18h after puparium formation, but exhibiting no obvious defects in the dendritic tree at the white pupa stage, and leads to no defects in the elimination of ddaF neurons by apoptosis at 16h after puparium formation, as compared to controls; at the wandering third instar larval stage, this expression also leads to dorsal multidendritic ddaC neurons exhibiting Golgi apparatus defects, as shown by severe decreases in the numbers of GalT or GM130 punctate structures in the soma, exhibiting decreased protein secretion, but exhibiting no obvious defects in ER exit sites, as shown by the apparently normal pattern of Sec31 punctate structures in the soma, as compared to controls.
