Inhibition (using Ctet\tetXTNT-LC.UAS) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT043656 and Hsap\RELAAD.VT040012 reduces the ability of males to orient towards and remain close to a female, compared to controls; they also lose the bias seen in controls to extend the wing closest to, rather than furthest from, the female; following of females, wing extensions and copulation are significantly reduced, compared to controls; courtship of decapitated (immobile) females is unaffected, compared to controls; this inhibition does not affect the ability of adult males to track a rotating grating or a bar, but they are less able to track small fly-sized objects, with no effect on the overall distribution of turning and walking speeds and no effect on movement in the absence of motion stimuli, compared to controls.
Feminization (using traUAS.cFa) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT043656 and Hsap\RELAAD.VT040012 in adult males does not affect tracking or courtship of females, compared to controls.
Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.20xUAS.FRT.Venus with Scer\FLP1G5.hs.Tag:PEST(mOdc) and 627 nm (red) light) of unilateral subsets of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT043656 and Hsap\RELAAD.VT040012 in adult males results in increased ipsilateral turning and significantly increased ipsilateral wing extensions, compared to controls, with no effect on walking speed.
Unilateral optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with 627 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT043656 and Hsap\RELAAD.VT040012 in adult males results in increased ipsilateral turning, compared to controls.
