Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change. The mutation is specific to the Galphaq-PD isoform.
GαqV303D homozygous as well as GαqV303D/Df(2R)Gαq1.3 or GαqV303D/Df(2R)Exel7121 transheterozygous adults show no electroretinogram (ERG) response to light stimulation. The GαqV303D mutants also display only a minimal response to a very bright illumination and their retinal tissue show rapid light-dependent degeneration (after six days in light/dark cycles the rhabdomeres in the mutant retina are largely missing or severely damaged), the ommatidial structure is intact in 1-day-old flies or flies kept in darkness.

The GαqV303D allele cannot be complemented by either Df(2R)Gαq1.3 or Df(2R)Exel7121 as they cannot rescue the flat electroretinogram (ERG) response to light stimulation observed in GαqV303D mutants, GαqV303D/Df(2R)BSC485 transheterozygotes show normal ERG response.
Scer\GAL4GMR.long-driven expression of Gαq.Scer\UAS (but not GαqV303D.Scer\UAS) can rescue the loss of response to light stimulation (flat electroretinogram, ERG, response) along with the rapid light-dependent retinal degeneration characteristic for GαqV303D adult flies. Expression of GαqV303I.Scer\UAS also rescues the lack of light response in ERG measurements.
