The potential native promoter of pnut (1.7kb upstream of the translation start site) drives expression of pnut cDNA sequence that carries mutations to mimic constitutive phosphorylation (amino acid replacements T509E and S517E). The open reading frame is tagged at the N-terminal end with Tag:FLAG.

      Combination with pnutT509E.S517E.T:Zzzz\FLAG rescues the larval lethality of pnutmut1 homozygous mutants and the emerging adult females are fertile but the rescued adult flies display significantly reduced viability at 25[o]C (but not at either 18[o]C or 29[o]C).
