Description

Exon 3 of endogenous sdt is replaced by in frame sfGFP. This results in sfGFP-tagged proteins lacking the sequence of exon 3, hence lacking the neur binding motif.

Stage 9/10 sdtΔ3-GFP embryos do not show any obvious defects in epithelial integrity or apical-basal cell polarity (assessed by the localization of aPKC and Nrt proteins). Stage 10 embryos show defects in posterior midgut remodeling, as shown by significantly fewer PGCs migrating through the posterior midgut epithelium, as compared to controls. In mutant eye discs, about half of ommatidia fail to form the expected six-cell rosettes and instead cells form elongated structures favoring early R3/R4 contacts, In adults, about half of ommatidia fail to form a rosette.
